Test principle || This kit was based on sandwich enzyme-linked immunosorbent assay technology. It was precoated with mouse anti-COL2 antibody on 96-well plates. And biotin-conjugated anti-mouse COL2 antibody was used as detection antibodies. Standards, test samples, and biotin-conjugated detection antibodies were subsequently added to the wells and washed with wash buffer.

HRP streptavidin was added and unbound conjugates were washed with wash buffer. TMB substrates were used to visualize the HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue product that changed to yellow after adding an acid stop solution. The density of yellow is proportional to the amount of mouse COL2 sample captured on the plate.
